Before you start

This course builds on Biology Fundamentals and Lab Techniques & Instrumentation. Take them first if you are new to the topic.

About this course

An intermediate course for future biochemists, biological technicians and medical laboratory technologists, about the core methods of a molecular biology lab and the data they produce. The first module covers nucleic acids: how DNA and RNA extraction works and how to judge yield and purity, how PCR works and how to check a primer pair, and how to compute relative gene expression from qPCR with the delta-delta-Ct method. The second module covers separation and detection: reading an agarose gel against a ladder, each step of SDS-PAGE and Western blotting, and calculating concentrations from an ELISA standard curve. The third module covers sequencing and bioinformatics: Sanger compared with next-generation sequencing, finding motifs and GC content in a DNA sequence with Python, and running a BLAST search and reading its results. Python activities are fill-in-the-blank and use only the standard library, so no programming background is needed. Every activity is a paper, data or web exercise: nothing here involves handling chemicals, biological material or patient samples at home. Bench work needs supervised training in a teaching or employer lab, and these careers have real entry requirements, such as a degree with lab coursework, and for medical laboratory scientists a NAACLS-accredited program and ASCP certification.
